I may not be understanding the question.

for index,letter in enumerate(s.lower()):

for index,letter in enumerate(s.lower()):
    if index==0:
        letter=letter.upper()
    print("{},{}".format(index,letter))

my_list=s[::-1]
for index,letter in enumerate(my_list.lower()):
    print("{},{}".format(index,letter))

i don't think you need enumerate here, just return (instead of print) a tuple with the four modifications of the given string. look at the title and capitalize methods also, they may help.
